چکیده

W e have isolated temperature resistant revertants from temperature sensitive E. coli strains containing either a thermolabile g lutaminyltRNA synthetase or leucyl-tRNA synthetase. Among the revertants which still contained the thermolabile leucyl-tRNA shynthetase w e found two classes of regulatory mutants (leuX and leuY) which have elevated levels of this enzyme. T h e leuX mutation specifies an operator-promoter region adjacent to the structural gene (leuS) for the enzyme. T h e leuY gene maps away from the leuS gene and codes for a protein. Us ing these mutants we demonstrated that the levels of leucyl-tRNA are related to the derepression of the leucine and isoleucine-valine operons. Among the revertants which still' contained the thermolabile g lutaminyltRNA synthetase we characterized three classes of mutants, glnT, glnU, and glnR. T h e glnT and glnU mutants contain elevated levels of t R N A 0 ' " , while the glnR mutant possesses elevated levels of glutaminyl tRNA synthetase. T h e level of glutamine synthetase, the enzyme responsible for the formation of glutamine, is also derepressèd in the gin T and glnR mutants.

Nanotoxicity for E. Coli and Characterization of Silver Quantum Dots Produced by Biosynthesis with Eichhornia crassipes

Nanomaterials are widely used in health and biomedical applications, however, only a few studies investigate their toxic effects.  The present report signifies a contribution to the study of the toxic effects of silver nanoparticles on   E. coli cells, which is a model organism of anthropogenic pollution. The toxicity of nanoparticles depends on their chemical and surface properties, shape and ...

Regulation of synthesis of the branched-chain amino acids and cognate aminoacyl-transfer ribonucleic acid synthetases of Escherichia coli: a common regulatory element.

Regulation of isoleucine, valine, and leucine biosynthesis and isoleucyl-, valyl-, and leucyl-transfer ribonucleic acid (tRNA) synthetase formation was examined in two mutant strains of Escherichia coli. One mutant was selected for growth resistance to the isoleucine analogue, ketomycin, and the other was selected for growth resistance to both trifluoroleucine and valine. Control of the synthes...

Purification of an Escherichia coli leucine suppressor transfer ribonucleic acid and its aminoacylation by the homologous leucyl-transfer ribonucleic acid synthetase.

The suppression of an amber mutation in a permissive strain of Escherichia coli can be achieved by a new tRNA species, a suppressor tRNA (for a review, see Reference 1). The tyrosine amber suppressor tRNA arises from a redundant tRNA species in the Sustrain by a single base change in the anticodon (2). However, this may not be the only mechanism to generate suppressor tRNA species, as suggested...

Regulation of synthesis of the aminoacyl-transfer ribonucleic acid synthetases for the branched-chain amino acids of Escherichia coli.

The regulation of synthesis of valyl-, leucyl-, and isoleucyl-transfer ribonucleic acid (tRNA) synthetases was examined in strains of Escherichia coli and Salmonella typhimurium. When valine and isoleucine were limiting growth, the rate of formation of valyl-tRNA synthetase was derepressed about sixfold; addition of these amino acids caused repression of synthesis of this enzyme. The rate of sy...
